Comparative bioinformatics analysis of acyl-CoA dehydrogenase from Galleria mellonella (Lepidoptera: Pyralidae)

Abstract:
Plastics are synthetic polymers derived from crude oil which can remain in the environment for many years due to high resistance to biodegradation. There are some reports on the ability of Galleria mellonella (Lepidoptera: Pyralidae) in degrading polyethylene. Previously it has been shown that biodegradation of polyethylene in this insect is associated with the function of several G. mellonella’s own enzymes, including acyl-CoA dehydrogenase. However, the genetic basis of this enzyme has not been described in detail. In this study, I used comparative bioinformatics to provide structural insight into acyl-CoA dehydrogenase. Genomic gmAcox1 has 2954 nucleotides encoding 1224 linear mRNA, five exons and four introns. GmAcox1 protein has 407 amino acids with a molecular weight of 43.8 kDa and a pI of 8.41. In addition, the total number of negatively charged residues (Asp + Glu) and positively charged residues (Arg + Lys) are 44 and 48, respectively. Using DnaSP analyses for five Acox1 genes from different species a total of 1002 variable (polymorphic) sites were observed 256 and 746 of which were singleton variable sites and parsimony informative, respectively. Variance of haplotype diversity is 0.01600 with standard deviation of haplotype diversity= 0.126. In addition, conserved regions are MWS MDS SKR RYR RDK SAK MWG MTG SKG SKG CAK CWW CYA CHR CHR MNR ANG VKG GTG RBR AWW RRD KKH YW in position 449-519 and MWG MTG SKG SKG CAK CWW CYAC HRC HRM NRA NGV KGG TGR BRA WW RRD KKH YWW WBR RYR SMA MAW RHT GKW KNAY HAM YRR YKR TKA NRR VAR WRM NGC HKY WGT VKT HKY WRC WAC in position 467-584. Tajima’s test indicated that Nucleotide diversity, Pi= 0.50694 and total number of mutations, Eta= 1286. MEGA6 software drawn phylogenetic tree indicated that G. mellonella Acox1 gene and Amyelois transitella are in one cluster. In addition, protein alignment indicated that Acox1 sequence in these two species have 90% identity, therefore, we suggest that biodegradation of polyethylene in Amyelois transitella should be considered in the future.
