Genetic diversity of Scolytus rugulosus (Muller, 1818) in Alborz mountain of Iran

Abstract:
Scolytus rugulous (Muller, 1818) is one of the most important bark beetles species that causes high damage to Orchard in the worldwide. The first step in control of bark beetles is accurate identification. To investigate the genetic diversity of Scolytus rugulous, during 2013-2016 sampling was done in six different elevation of Alborz Mountain on Prunus sp.. The sampling method through direct observation was done by brush and knife. In each elevation, 16 individuals were collected in different area and identified morphologically. In the present study CO1 region of mitochondrial DNA was used to analyze the genetic diversity. The DNA extraction was based on the Salting out method. PCR products were sequenced and aligned by MEGA ver.07. The AMOVA analysis was done by Alrequin ver 0.3 software. The result revealed significant differences between populations in different area. Thirteen haplotypes were identified which were clustered in two different groups. The Fst index was calculated as 0.6 which showed high genetic diversity between these populations. The result demonstrated that there were two different populations of Scolytus rugulosus in two different area of Alborz Mountain. According to findings of the present study, the Alborz Mountain is considered as a geographical barrier and, therefore, management of Scolytus rugulosus may need different control methods.
